Η έκθεση, οργανωμένη κυρίως χρονολογικά, παρουσιάζει την ιστορία της Τήλου από τους προϊστορικούς χρόνους (4.000 π.Χ.) έως τους νεότερους (17ος αι.). Από τα απολιθώματα των νάνων ελεφάντων και τα πρώτα ίχνη ανθρώπινης κατοίκησης έως τις τοιχογραφίες των βυζαντινών και μεταβυζαντινών ναών και τα νεότερα κειμήλια, τα εκθέματα φέρνουν τον επισκέπτη σε επαφή με τη φυσιογνωμία του νησιού στο πέρασμα του χρόνου.
